How to evaluate emerging vitamin D and COVID-19 intervention trials | Roger Seheult

Vitamin D serves as a major regulator of immune function. Meta-analyses of randomized controlled trials show that daily or weekly vitamin D supplementation — but not large bolus doses — protects against acute respiratory tract infections. In the context of COVID-19, the authors of a recent double-blind, placebo-controlled trial concluded that hospitalized COVID-19 patients administered a single large vitamin D3 dose did not have statistically better clinical outcomes, as measured by hospital length of stay, than those given a placebo. The authors acknowledge several limitations to their study, including a patient population with several comorbidities, multiple medications, and lacking vitamin D deficiency. Moreover, the sample size may be too small to detect modest, yet clinically meaningful differences between the vitamin D3 and placebo groups.
